Biotic analysis of the implementation of standard operating procedures (SOP) in banana tissue culture in the UPTD seed laboratory

Abstract
Banana tissue culture is an aseptic plant propagation technique that plays a crucial role in producing superior, uniform, and disease-free seedlings. The success of this technique is greatly influenced by the implementation of Standard Operating Procedures (SOPs) and the application of bioethical principles in laboratory activities. This study aims to analyze the implementation of banana tissue culture SOPs and their compliance with bioethical principles in the Horticultural Parent Seed UPTD Laboratory. The study used a qualitative approach with descriptive-analytical methods. Data were obtained through direct observation, interviews, and documentation during laboratory activities. The results showed that the implementation of SOPs has not been optimal, indicated by the non-mandatory use of personal protective equipment (PPE) during media preparation, the transfer of equipment through open areas that have the potential to cause contamination, and limited laboratory facilities, especially water availability. From a bioethical perspective, these conditions indicate a suboptimal implementation of the principles of occupational safety, scientific responsibility, and laboratory sterilization. Therefore, increased supervision, disciplined implementation of SOPs, and improvements to laboratory facilities are needed to support safe, sterile, and bioethical tissue culture activities.
